SCOPE The Doudna lab at the Gladstone Institutes is focused on delivering genome editing enzymes to specific cells in the body. We are working on projects to make it easier to deliver tools that can edit genes into cells, especially outside the liver. To do this, we want to create special particles inspired by viruses to carry these tools into immune cells. As a Research Intern, you will help test how different parts of these virally-derived particles affect their ability to edit genes in cells. SCOPE A main goal of the HEAL fellowship is to recruit a cohort of fellows and support the transformation of our fellows and alumni to lead in the global health equity movement. Our two large multi-day intensive summer training events with health professionals from all over the world supports this goal. The interns are required to travel, attend, and support the logistics of these training sessions in both Flagstaff, Arizona, and in Santa Cruz, California. The summer training sessions will last about three weeks away from home (June 20-28 and July 7-19). All travel expenses will be covered directly by HEAL. Recently, a new information-gathering method called “trogocytosis” has been discovered that allows APCs to sample parts of a cell without destroying it. This process presents promising directions to understand disease. On one hand, this process may enable the immune system to monitor cells without damaging healthy tissue. On the other hand, trogocytosis may be exploited by diseased cells, like cancer cells, to avoid destruction. Over the course of 10 weeks, you will use genetic engineering to identify proteins expressed by APCs that regulate trogocytosis. Specifically, you will be executing a CRISPR screen under the supervision and guidance of a postdoctoral fellow with additional support from other Krummel lab members and UCSF collaborators. This will include the creation of the CRISPR library, the performance of the genetic screen, preparation of genomic DNA for sequencing, and analysis of the data. By the end of your internship, we aim to have a list of candidate genes that either promote or inhibit trogocytosis. The mechanisms revealed by our investigation will provide important insights into how the immune system monitors healthy tissues and protects them from disease throughout our lives. SCOPE We are interested in understanding how the many components of the immune system come together to fight cancer. The immune system is made up of multiple cell types: those that can kill cancer cells, some that protect healthy cells, and others that train other immune cells to identify what is healthy and not. SCOPE Our laboratory studies a lung blood vessel disease called pulmonary hypertension. We use mouse models of the disease, and compare our results to human data. The specific focus of our laboratory is how the immune system contributes to pulmonary hypertension by inflammation causing lung blood vessel injury. The position would be for a member of our laboratory, performing standard molecular biology techniques, and helping with mouse handling, breeding and sample collection.
